Thermocyclers, or polymerase chain reaction (PCR) machines, are laboratory instruments that are used to amplify DNA and RNA samples by the PCR by regulating temperature during cyclical programs. The PCR is considered as one of the most important developments in molecular biology. Real-time PCR is performed in a thermal cycler with the conventional technique of measuring fluorescence. A wide range of fluorescent chemistries are employed to monitor the PCR in progress. Thermal cyclers find applications mostly in research laboratories, where they are mainly used for amplifying DNA, for cloning DNA, and for sequencing genes. They are also employed in diagnosing certain diseases and genetic disorders and analysis of microbial communities. The development of PCR-based genetic or DNA fingerprinting protocols has resulted in their widespread application in forensics.
